Specimen:

Plasma from immunocompromised patients – 1mL required (minimum 300 uL). CSF from encephalitis cases – 600uL required (minimum 300uL). Minimum volume will not allow repeat testing of samples with equivocal results.

Collection Method:

Collect blood in plasma preparation tube (PPT) or K2EDTA tube (plastic; lavender or pink top). Do not use heparin as an anti-coagulant. Centrifuge tube at room temperature at 1,100 RCF for 10-15 min. Aliquot plasma into separate tube. Collect CSF in a dry tube subject to classic conditions for carrying out lumbar punctures.

Specimen Processing, Storage and Shipping:

Specimens may be stored and shipped refrigerated to NML within 48h of sample collection or must be stored and shipped frozen.

Transportation of Dangerous Goods:

Shipping of specimens shall be done by a TDG certified individual in accordance with TDG regulations. For additional information regarding classification of specimens for the purposes of shipping, consult either Part 2 Appendix 3 of the TDG Regulations or section 3.6.2 of the IATA Dangerous Goods Regulations as applicable.

Patient Criteria:

Specimens are accepted from immunocompromised patients with primary infection or reactivation or patients with encephalitis.
** Please note that specimens for routine monitoring of HHV-6 infection in transplant patients will not be accepted.

Accompanying Documentation:

Completed "Viral STI, Polyoma and Herpesvirus Testing" requisition form including sender lab name, address and telephone number, patient identifier, date of birth or age and sex, specimen reference #, type of specimen, date collected, test requested, reason for testing and any other relevant clinical information.

Comments:

If investigation of HHV-6 chromosomal integration is required, please contact lab for further details prior to specimen collection.

Methods and Interpretation of Results:

DNA is extracted from specimens and tested by real-time PCR using the PrimerDesign Quantification of HHV-6 genesig kit (1), which targets the BHLF3 putative dUTPAse gene. The results are reported as copy number of HHV-6 DNA per mL of original specimen. This assay is for research purposes only.
